Baseado na clássica história infantil do Dr. Seuss, Horton Hears a Who! conta a história de Horton, um elefante amistoso, que ouve um dia um grito de socorro vindo de um grãozinho de poeira. Investigando mais de perto, descobre toda uma raça de "Whos" microscópicos a viverem sobre ele. Horton torna-se amigo dos Whos e protege o seu mundo minúsculo, mas os seus vizinhos da selva temem que ele tenha enlouquecido, acreditando em coisas que não existem. Presumindo que nada tão pequeno pode possivelmente estar vivo, os amigos de Horton conspiram para destruir o grãozinho de poeira de modo a fazê-lo sair das suas alucinações, mas ele está decidido a salvar a partícula minúscula.
